Test pLsrR/cI

From GcatWiki

pLsrA/cI is constitutively on and is repressed by LsrR. The addition of AI-2, which becomes phosphorylated by LsrK, represses LsrR causing the promoter to turn on. The addition of cI to should repress the promoter causing it to turn off.

1)Test LsrA repression.

Some promoter→RBS+LsrR+TT+pLsrA/cI→GFP does not glow

. B0034+K091001+B0015+K091106→E0240

add AI-2

Some promoter→RBS+LsrR+TT+pLsrA/cI→GFP glows

. B0034+K091001+B0015+K091106→E0240

2)Test cI repression.

Some promoter→RBS+cI+TT+pLsrA/cI→GFP does not glow

. B0034+C0051+B0015+K091106→E0240

add cI

Some promoter→RBS+cI+TT+pLsrA/cI→GFP does not glow

. B0034+C0051+B0015+K091106→E0240

3)Test LsrA and cI repression.

Some promoter→RBS+LsrR+cI+TT+pLsrR/cI→GFP does not glow

. B0034+K091001+C0051+B0015+K091106→E0240

add AI-2

Some promoter→RBS+LsrR+cI+TT+pLsrR/cI→GFP does not glow

. B0034+K091001+C0051+B0015+K091106→E0240

4)Control.

pLsrR/cI→GFP glows

K091106→E0240
